CW January 2006Issue Number 1 January 2006 ISSN 10593802 Among Imatinib Resistant/Intolerant Patients, Responses to Dasatinib in Advanced CMLIn vivo, dasatinib (BMS-354825) is several hundred-fold times more active than imatinib (Gleevec, Novartis) and is non-cross resistant with imatinib. It is fully active against 18/19 imatinib-resistant BCR-ABL mutants in vivo, and overcomes resistance conferred by SRC family kinase activation. A Phase II clinical trial of the new, oral, multi-targeted kinase inhibitor among patients with imatinib-resistant or -intolerant chronic myeloid leukemia (CML) showed that within the first six months, 90% of patients reached a complete hematologic response, 45% achieved a major cytogenetic response, and 33% achieved a complete cytogenetic response. Two further studies among patients with more advanced disease (accelerated phase or blastic phase) continue to show impressive responses with the new agent, according to a presentation at the 147th Scientific Sessions and Exposition of the American Society of Hematology.
