CW September 2004Issue Number 9 September 2004 ISSN 10593802 Advanced Colorectal Cancer: Higher Response Rates for Iressa Added to FOLFOX-4Historical response rates with FOLFOX-4 (oxaliplatin 85 mg/m2, leucovorin 200 mg, 5-FU 400 mg/600 mg) among patients with untreated colorectal cancer have been 30-55%. They are lower (9%) in patients refractory to CPT-11 (irinotecan)/FU/LV (fluorouracil/leucovorin). Gefitinib (ZD1839, Iressa™, AstraZeneca), is an orally active, selective EGFR-TKI (epidermal growth factor receptor tyrosine kinase inhibitor) which blocks signal transduction pathways implicated in tumor cell proliferation and cell survival. In preclinical studies, it has shown supra-additive anti-tumor activity in combination with a number of chemotherapy classes, including platinums.
