Oncogenes as Potential Targets for Chemotherapy

Raf kinase, an enzyme that acts in the Ras signaling pathway, is involved in cancerous cell growth. Thus, small molecule inhibitors of Raf kinase activity may be important agents for the treatment of cancer. A novel class of Raf kinase inhibitors was discovered, using a combination of medicinal and combinatorial chemistry approaches. This effort culminated in the identification of a compound, BAY 43-9006, which is currently undergoing phase I clinical trials and shows promising results in the treatment of melanoma and other cancers.
