5-FU Still Reigns Supreme?

For more than 40 years, 5-fluorouracil (5-FU) has been considered the most effective drug in the treatment of colorectal cancer (CRC). While the majority of chemotherapeutic agents for other cancers continuously undergoes changes and improvements, 5-FU has remained firmly entrenched. It is extremely toxic and never curative, but only palliative at best. Although CRC is a frequent malignancy, surgery and radiation are basically the most effective treatment from the patients? point of view, and there is disappointingly little progress in novel chemotherapy, which is tolerable to the patients.
