G-Quadruplex and Oncogene Activation

The G-quadruplex structure at the major promoter site of c-MYC gene is destabilized by a single base mutation that increases its transcriptional activity by 3-fold over the wild type gene. Small molecules that interact with it and prevent transcription can achieve stabilization of this tetrad structure.
