CW August 2002Issue Number 8 August 2002 ISSN 10593802 Pemetrexed: a New AntifolateThat Pemetrexed is considered a novel agent is characteristic for the tardiness in testing and accepting promising compounds: the drug has been described in 1992 and has entered Phase II evaluation only recently. It is broadly active in various solid tumors and demonstrates excellent synergy in combination with other agents, such as with Gemcitabine. Pemetrexed is a multitargeted antimetabolite able to inhibit at least three folate-dependent enzymes and is well tolerated.
