CW August 2001Issue Number 8 August 2001 ISSN 10593802 Antiestrogens that Circumvent Tamoxifen Resistance in Breast CancerThe use of Tamoxifen in the adjuvant therapy of breast and other cancers is in its third decade, testimony to its benefits and popularity. There are still many papers being published on its effect ? and that of other non-steroidal antiestrogens. Apart from the hotly debated value in cancer chemoprevention, the duration of Tamoxifen administration is a much-discussed topic; a consensus for an optimal time period for the effectiveness of Tamoxifen has not yet been reached. However, drug resistance, the ultimate defense mechanism of malignant cells, raises its ugly head sooner or later. So far, the search for second or even third line antiestrogenic agents is only moderately successful; the recent introduction of GW5638 is being followed with hope and interest.
