Conjugation of paclitaxel with polyglutamate renders it easily available to tumor cells than normal cells because blood vessels supplying the tumors are more porous to the conjugate than the blood vessels supplying the normal tissues. Additionally, a proteolytic enzyme cathepsin B is present in significant amount in rapidly growing tumors that releases active paclitaxel from its conjugate in the tumor over a period of time maximizing its effect.
